ESV - 29 Jesus answered them, "This is the work of God, that you believe in him whom he has sent.

In the context of the verse cited in the question, Jesus was addressing those who had sought after Him because He had recently miraculously fed a large crowd of people from a limited amount of food. Jesus told them not to work for food that spoils, but for the food that would lead to eternal life that only He could give them -- that "food" being faith in Him. He then elaborated on that point by specifically saying (as noted in the cited verse) that the "work" that the people could perform was thus believing in Him by responding affirmatively to the Holy Spirit's prompting. (Although "faith" and "works" are often viewed in contrast to one another, this verse thus shows how they are interrelated.) Believers are then to "work" (through the power of the Holy Spirit) to bring others to the same saving faith.
